LCMS-B: rt 3.20 min, m/z = 356.8 [M+Na]+, 334.8 [M+H]+.

Solid sodium carbonate was added until a pH of 10 was reached, and the mixture was stirred at ambient temperature for 1 hour. The organic layer was separated and the aqueous layer back-extracted withdichloromethane (4 x 30 ml_). The combined organic layers were passed through a phase separator column and concentrated under reduced pressured to yield crude ethyl 3-(1 ,2-benzoxazol-3-ylamino)-3- oxopropanoate.

The residue was purified by silica gel column chromatography using hexane/ethyl acetate (4:1) as solvent system. Fractions containing earlier eluted product were combined and concentrated to obtain l-[3-(6-chloro)-benzisoxazoyl]-5- trifluoromethoxylbenzimidazole-2-one as a white solid. 1H NMR (DMSO, 500 MHz) delta 8.28 (d, J=9.0Hz, IH), 8.12 (s, IH), 7.74 (d, J=9.0Hz, IH), 7.54 (d, J=8.5, IH), 7.14 (d, J=8.5, IH), 7.13 (s, IH). Fractions containing later eluted product were combined and concentrated to obtain l-[3-(6-chloro)-benzisoxazoyl]-6-trifluoromethoxylbenzimidazole-2-one as a white solid. 1H NMR (DMSO, 500 MHz) delta 8.28 (d, J=9.0Hz, IH), 8.12 (s, IH), 7.74 (d, J=9.0Hz, IH), 7.66 (brs, IH), 7.54 (d, J=8.5, IH), 7.13 (brs, IH).

The reaction mixture was stirred overnight at RT. The mixture was washed 2 times with H20. The organic layer was separated, dried (MgS04), filtered off and the solvent was evaporated. The residue was purified by column chromatography over silicagel (eluent CH2Cl2) yielding O. OlOg compound 9 (yield of 2%, Melting Point 133C) and a fraction which was further purified by column chromatography over silicagel (eluent hexane/CH2Cl2) yielding 0.225 g of compound 10 (yield of 38%, Melting Point 97C-100C), and 0.100 g of compound 11 (yield of 17%, Melting Point 154C-160C).

Benzisoxazole-3-amine 2 (2.68 g, 20 mmol) was dissolved in water (25 ml) and concentrated HCl (12.5 ml) under vigorous stirring in an ice/water bath. A freshly prepared, ice-cold solution of NaNO2 (1.4 g, 20 mmol) in water (10 ml) was added drop wise to the reaction mixture by keeping the internal temperature between 0 to 5 C. After the completion of addition, the reaction mixture was stirred for an additional 10 min. A freshly prepared solution of sodium azide (1.3 g, 20 mmol) in water (10 ml) was added drop wise to the reaction mixture via an additional funnel while keeping the internal temperature of the reaction mixture below 5 C. Upon complete addition of the sodium azide solution, the reaction mixture was stirred for an additional 20-30 min at 0 C, followed by stirring at room temperature for another 3 h. The reaction mixture was extracted with ethyl acetate EtOAc (25 ml) three times. The combined organic layer was washed with water (25 ml) and brine (25 ml), dried over anhydrous sodium sulphate and concentrated under reduced pressure to get crude benzisoxazole-3-azide 3, which was purified by column chromatography over silica gel (60-120 mesh) using hexane/EtOAc (8:2) as eluent. The pure benzisoxazole-3-azide 3 was stored at 2-5 C in the refrigerator.

Step 1: To a -78 C solution of ((S)-2-oxo-2,3,4,5-tetrahydro-1H-benzo[bl[1,4ldiazepin- 3-yl)-carbamic acid tert-butyl ester (807 mg, 2.91 mmol) in THF (19.4 mL) was added lithium bis(trimethylsilyl)amide (3.2 mL, 1.0 M solution in THF, 3.2 mmol), dropwise. The mixture was stirred at -78 C for 15 mm, then a mixture of Nal (523 mg, 3.49 mmol) and 3- (bromomethyl)benzo[dlisoxazole (740 mg, 3.49 mmol) in THF (9.7 mL) was added dropwise over 10 mm. The mixture was stirred at -78 C for 50 mm., warmed to RT and stirred for 4.5 h. The mixture was diluted with 1 N citric acid and extracted with EtOAc. The combined extractswere washed with sat. aq. NaHCO3, brine, dried over Na2SO4, filtered, and concentrated. The residue was purified by flash chromatography. The resulting material was repurified by flash chromatography to provide a -4:1 mixture of (S)-tert-butyl 1-(benzo[dlisoxazol-3-ylmethyl)-2- oxo-2,3 ,4,5-tetrahydro- 1 H-benzo [bI [1 ,4jdiazepin-3-ylcarbamate and (S)-tert-butyl 1- (benzo [djisoxazol-3 -ylmethyl)-4-oxo-2,3 ,4,5 -tetrahydro- 1 H-benzo [bI [1,41 diazepin-3-ylcarbamate (742 mg, 63 %) as a yellow foam. LC-MS mlz 431 [M+Naj.Step 1: To a 0C solution of -4:1 mixture of tert-butyl (S)-1-((S)-1-(benzo[djisoxazol-3- ylmethyl)-2-oxo-2,3 ,4,5-tetrahydro- 1 H-benzo [bI [1,41 diazepin-3 -ylamino)- 1 -oxopropan-2- yl(methyl)carbamate and tert-butyl (S)-i -((S)-i -(benzo[dlisoxazol-3-ylmethyl)-4-oxo-2,3 ,4,5- tetrahydro- 1 H-benzo [bI [1 ,4ldiazepin-3-ylamino)- 1 -oxopropan-2-yl(methyl)carbamate (209.4 mg, 424 tmol) in CH2C12 (4.24 mL) was added pyridine (343 tL, 4.24 mmol), followed by methyl 4-(chlorocarbonyl)benzoate (92.7 mg, 467 tmol). After 1 h at 0 C the mixture was diluted with H20 and extracted with CH2C12. The combined extracts were washed with 1 N aq. citric acid, sat. aq. NaHCO3, and brine, dried over Na2504, filtered, and concentrated. The residue was purified by flash chromatography. The resulting material was repurified by flash chromatography to provide methyl 4-((S)-5-(benzo[djisoxazol-3 -ylmethyl)-3 -((S)-2-(tert-butoxycarbonyl(methyl)amino) propanamido)-4-oxo-2,3 ,4,5 -tetrahydro- 1 Hbenzo[b l[1,4ldiazepine-1-carbonyl)benzoate (194 mg, 70 %) as a white solid. Benzoic isoxazol-3-bis(N,N-t-butoxycarbonyl)amine (122): To a solution of 121 (148 mg, 1 mmol) in DCM (2 mL) was added di-t-butyldicarbonate (546 mg, 2.5 mmol), DIEA (0.348 mL, 2 mmol), and DMAP (22.2 mg, 1 mmol) at O0C. The reaction was warmed up to RT and stirred for 4 hr and then diluted with ethyl acetate and treated with aqueous ammonium chloride to pH 6. The organic layer was separated and washed with brine and concentrated in vacuo. The residue purified by silica gel chromatography using 100% hexane as the eluent to yield 122 (347 mg, 100%), LC-MS (ESI) m/z 348.8.

651780-27-7, To a stirred solution of ethyl 6-bromo-1,2-benzoxazole-3-carboxylate (1.1 g) in THF (10 ml) and EtOH (3 ml), LiBH4 (0.133 g) was added drop wise at 0oC under inert atmosphere. The re- action mixture was stirred at 0oC for 2 h. Reaction was monitored by TLC. After the completion, the reaction mixture was diluted with Water (20 ml), quenched with 5% NaOH solution and ex- tracted with Ethyl acetate (30 ml x 2). The combined organic extracts were dried over Na2SO4, filtered and concentrated under reduced pressure. The crude product was purified by flash chro- matography (eluting in 15-20% ethyl acetate in heptane as a mobile phase) to afford (0.8 g) of the title compound. HPLC/MS (method 1): Rt : 1.587 min; MS: m / z = 229 (M+1).

To a stirred solution of 1-(3-phenyl-1,2,4-thiadiazol-5-yl)piperidine- 4-carboxylic acid (289 mg, 1.00 mmol) and DMF (0.010 mL) in THF (5.0 mL) was added oxalyl chloride (0.174 mL, 2.00 mmol) dropwise at 0 C. After stirring at room temperature for 1 h, the solvent was concentrated in vacuo. To the residue were added THF (5.0 mL), benzo[d]isoxazol-3-amine (134 mg, 1.00 mmol), and pyridine (0.404 mL, 5.00 mmol) at 0 C. The mixture was stirred at room temperature for 1.0 h, and the solvent was concentrated in vacuo. The residue was purified by silica gel column chromatography (hexane-EtOAc), and recrystallized from EtOAc- hexane to give 65a (60.3 mg, 15%) as colorless crystals, mp 204- 205 C. 1H NMR (300 MHz, CDCl3) d: 2.05-2.22 (4H, m), 2.88 (1H, bs), 3.36-3.45 (2H, m), 4.11-4.18 (2H, m), 7.32-7.63 (6H, m), 8.19-8.27 (3H, m), 9.46 (1H, br s). Analytical HPLC showed 97.4% purity.
