Heteroaryl urea inhibitors of fatty acid amide hydrolase: Structure-mutagenicity relationships for arylamine metabolites

The structure-activity relationships for a series of heteroaryl urea inhibitors of fatty acid amide hydrolase (FAAH) are described. Members of this class of inhibitors have been shown to inactivate FAAH by covalent modification of an active site serine with subsequent release of an aromatic amine from the urea electrophile. Systematic Ames II testing guided the optimization of urea substituents by defining the structure-mutagenicity relationships for the released aromatic amine metabolites. Potent FAAH inhibitors were identified having heteroaryl amine leaving groups that were non-mutagenic in the Ames II assay.

Fumarate hydratase (fumarase), a vulnerable component of the citric acid cycle in Mycobacterium tuberculosis (Mtb), is a metabolic target that could satisfy this unmet demand. A key challenge in the targeting of Mtb fumarase is its similarity to the human homolog, which shares an identical active site. A potential solution to this selectivity problem was previously found in a high-throughput screening hit that binds in a nonconserved allosteric site. In this work, a structure-activity relationship study was carried out with the determination of further structural biology on the lead series, affording derivatives with sub-micromolar inhibition. Further, the screening of this series against Mtb in vitro identified compounds with potent minimum inhibitory concentrations.

The present invention describes an improved process for the preparation of 1,2Benzisoxazole-3-methanesulphonamide (Zonisamide), a well known anti-epileptic agent which possesses anti-convulsant and anti-neurotoxic effects. Other aspect of this invention are isolation of a key intermediate, viz., crystalline sodium chloride associated 1,2Benzisoxazole-3-methane sodium sulfonate (BOS-Na:NaCl). The isolated crude BOSNa:NaCl is directly converted to Zonisamide using controlled molar ratio of chlorosulfonic acid to avoid the disulfonated side products. Different peripheral substituted phthalocyanines: Synthesis, characterization, aggregation behavior, antioxidant and antibacterial activity

In this study, a novel phthalonitrile, 4-chloro-5-(2-((2-hydroxyethyl)(p-tolyl)amino)ethoxy)phthalonitrile (3), and its metallophthalocyanine derivatives (4-6) are prepared by cyclotetramerization with appropriate metal salts in dimethylformamide. The newly prepared compounds have been characterized by several spectroscopic techniques. All compounds are evaluated for their antioxidant and antibacterial potential. For the antioxidant studies, three tests are applied; DPPH (2,2-diphenyl-1-picrylhydrazylradical) scavenging, metal chelating and reducing power activity. Compound 4 exhibits the best DPPH scavenging activity as 35.2% at 100 mg/L concentration. The metal chelating activities of compounds 3 and 4 are 69.7% and 56.4%, respectively. Reducing power activities of compounds 3 and 4 are higher than alpha-tocopherol which is used as positive control. All compounds show moderate antibacterial activity when compared to the standard antibiotics, amikacin and tetracycline.
