Polymorphic and solvate structures of ethyl ester and carboxylic acid derivatives of WIN 61893 analogue and their stability in solution

3-Ethyl ester- (1) and 3-carboxylic acid-isoxazole (2) derivatives of an antiviral drug analogue WIN 61893 were synthesized and characterized by X-ray crystallography and NMR spectroscopy. Crystallization experiments afforded two polymorphic structures for the ethyl ester derivative and two solvate structures for the carboxylic acid derivative based on their ability to form intermolecular hydrogen bonding interactions with the solvent molecules. The conformations of the derivatives depended greatly on the orientation of the planar isoxazole and phenyl-oxadiazole ring systems with respect to one another and were found to take up perpendicular, linear or tilted conformations. The carboxylic acid derivative was furthermore observed to undergo isoxazole ring cleavage by decarboxylation in DMSO solution and transforming into a beta-keto nitrile ring-opening product over several days, whereas, the isoxazole ring of the ethyl ester derivative was not affected.

Novel classes of 3-(6-Aryl-5-phenylsulfanylimidazo[2,1-b][1,3,4]thiadiazol-2-yl-methyl)-benzo[d]isoxazoles (2a-f) and 3-(5-Benzenesulfonyl-6-arylimidazo[2,1-b][1,3,4] thiadiazol-2-yl-methyl)-benzo[d]isoxazoles (3a-f) have been synthesized. The sulfide derivatives (2a-f) were obtained by the nucleophilic substitution of bromo derivatives (1a-f) with thiophenols, and sulfone derivatives (3a-f) were obtained by the oxidation of 2a-f. All the newly synthesized compounds were characterized by elemental analysis, infrared, nuclear magnetic resonance, and mass spectroscopic data. Furthermore, these novel sulfide and sulfone derivatives were screened for their antibacterial and antifungal activities against various microorganisms.

A Practical Electrophilic Nitrogen Source for the Synthesis of Chiral Primary Amines by Copper-Catalyzed Hydroamination

A mild and practical method for the catalytic installation of the amino group across alkenes and alkynes has long been recognized as a significant challenge in synthetic chemistry. As the direct hydroamination of olefins using ammonia requires harsh conditions, the development of suitable electrophilic aminating reagents for formal hydroamination methods is of importance. Herein, we describe the use of 1,2-benzisoxazole as a practical electrophilic primary amine source. Using this heterocycle as a new amino group delivery agent, a mild and general protocol for the copper-hydride-catalyzed hydroamination of alkenes and alkynes to form primary amines was developed. This method provides access to a broad range of chiral alpha-branched primary amines and linear primary amines, as demonstrated by the efficient synthesis of the antiretroviral drug maraviroc and the formal synthesis of several other pharmaceutical agents.

Substituent effect on the reductive N-dearylation of 3-(indol-1-yl)-1,2-benzisoxazoles by rat liver microsomes

The reductive metabolism of a series of 3-(indol-1-yl)-1,2-benzisoxazoles was examined in vitro using rat liver microsomes. 3-(Indol-1-yl)-1,2-benzisoxazole was reduced to the corresponding amidine (resulting from N-O bond cleavage) under anaerobic conditions. The reaction required viable microsomes and NADPH and was inhibited by carbon monoxide, air, and ketoconazole, suggesting the involvement of cytochrome P450 enzymes. The amidine was subsequently nonenzymatically hydrolyzed to 1-salicylindole, which in turn was hydrolyzed to indole. Addition of electron-withdrawing substituents (Cl-, MeSO2-) at the 6-position of the benzisoxazole ring resulted in a significant increase in the rate of substrate reduction. Introduction of electron-withdrawing substituents on the indole ring likewise increased the rate of substrate consumption but caused a substituent-dependent shift of the site of bond cleavage from the 1,2-isoxazole N-O bond to the C-N bond linking the 1,2-benzisoxazole to the indole moiety. In the case of 3-(2-chloro-3-methanesulfoxylindol-1-yl)-1,2-benzisoxazole, C-N bond cleavage was nearly quantitative, and products resulting from N-O bond reduction were not observed. The overall rates of 3-(indol-1-yl)-1,2-benzisoxazoles reduction were found to be substrate concentration-dependent and observed Michaelis-Menten-type behavior. The apparent V-max of substrate reduction by rat liver microsomes correlated negatively with the free energy of the lowest unoccupied molecular orbitals (E-LUMO) calculated semiempirically using a parameterized model 3 (PM3), and suggested that the initial electron transfer was rate-determining and that the E-LUMO could be used as an indication of the susceptibility of 1,2-isoxazoles to undergo reductive metabolism.

Paliperidone (9-hydroxyrisperidone), the primary active metabolite of risperidone, is a benzisoxazole derivative of the second-generation antipsychotics. Paliperidone patmitate or PP (the patmitate ester of paliperidone) is available as a once-daily oral tablet, a once-monthly injection or PP1M, and a 3-monthly injection or PP3M (marketed under the name of Trevicta (R) in the European Union). Despite the recent availability of PP3M, several cases of intoxication have been reported worldwide, including lethal cases. However, in Europe, no case of fatal poising has been reported to date. A 33-year-old man was found dead in his room at his parents’ home. His only medical background was a history of schizophrenia. The external examination was without any particularity except a runny nose of brownish liquid with an alcoholic smell. Given the context and the tack of evidence of a third party intervention, an autopsy was not considered necessary. Toxicological analysis of right femoral blood highlighted the presence of paliperidone (240 mu g/ L) only. The risk associated with the use of PP3M formulation is still imperfectly evaluated and several cases of intoxication have been reported worldwide, including fatal cases. In this case, paliperidone poisoning appears to be the highly likely cause of death. In fact, the blood concentration of paliperidone is high, twice the toxicity threshold used in therapeutic monitoring, and no other substances have been identified. This case highlights: the necessity to always evaluate the benefit-risk balance when prescribing this sustained-release form of paliperidone and the difficulties in interpreting paliperidone concentration in forensic cases. SYNTHESIS AND CATALYTIC PROPERTIES OF NON-CROSS-LINKED AND CROSS-LINKED POLY(ALKYLMETHYLDIALLYLAMMONIUM BROMIDES) HAVING DECYL, OCTYL, AND HEXYL SIDE-CHAINS

A family of non-cross-linked and cross-linked copolymers containing decyl, octyl, and hexyl groups as side chains ((CL)-CopolC1-10, (CL)-CopolC1-8, and (CL)-CopolC1-6, respectively) were synthesized by radical-initiated cyclocopolymerization of alkylmethyldiallylammonium bromide monomers without and with a small amount of N,N’-methylenebisacrylamide as a cross-linking agent in aqueous solution. Their H-1 NMR and IR spectra indicated the presence of five-membered rings cross-linked without and with N,N’-methylenebisacrylamide in the macromolecules. Viscosity measurements showed that the cross-linked copolymers exhibit a larger reduced viscosity in aqueous solution with increasing cross-linking agent content in the copolymers. For(CL)-CopolC1-10, the conformational transition to compact coils was indicated by changes of the reduced viscosity in dilute aqueous solutions. At higher concentrations, intermolecular aggregation was also revealed and increased with increasing the percentage of cross-linking for CL-CopolC1-10. (CL)-CopolC1-8 and (CL)-CopolC1-6 showed extented molecular dimensions in aqueous solution. The hydrophobic microdomains of the non-cross-linked and cross-linked copolymers were probed by hypsochromic shifts of the long-wavelength absorption band of Methyl Orange as a solvatochromic agent, noncovalently bound to the macromolecule. The unimolecular decarboxylation of 6-nitrobenzisox-azole-3-carboxylate (6-NBIC), catalyzed by these copolymers in aqueous solution, was used as a model reaction to study the influence of polysoap microenviroment on reactivity. Depending on the hydrophobic group content, (CL)-CopolC1-10 led to a remarkably large rate enhancement, whereas (CL)-CopolC1-8 induced only modest rate acceleration for the decarboxylation of 6-NBIC. A small rate enhancement was observed in the presence of(CL)-CopolC1-6. The decarboxylation rate is also sensitive to changes of the percentage of cross-linking in the macromolecules. A maximum in rate constant was found at about 0.2% (w/w) cross-linking agent for CL-CopolC1-10 and at about 0.4% (w/w) for CL-CopolC1-8 in plots of the rate constant vs cross-linking agent content.

Gradient selected gHMQC-TOCSY spectra of phenanthro[9,10-d][1,3]oxazoles 11-14 were obtained at different mixing times (12,24,36,48 and 80 ms) to identify the spin system where the protons of phenanthrene ring at H-5, H-6 and at H-9 and H-7 and H-8 were highly overlapping.
