Monitoring of zonisamide in human breast milk and maternal plasma by solid-phase extraction HPLC method

An HPLC method was developed for the determination of zonisamide in human breast milk and plasma. Chromatographic separation was achieved using a Develosil CN analytical column with potassium dihydrogenphosphate buffer (pH 3.5 with milk, pH 2.5 with plasma)-acetonitrile as the mobile phase. Zonisamide and 1,2-benzisoxazole-3-methansulfonamine acetate as internal standard were detected by ultraviolet absorbance at 240 nm. Zonisamide in breast milk and plasma was extracted by a rapid and simple procedure based on C-18 bonded-phase extraction. Determination of zonisamide in human breast milk and plasma was possible in the concentration range 0.05-20.0 mu g/mL. The recoveries of zonisamide added to human breast milk and plasma were 79.5-85.0% and 86.3-93.1%, respectively, with coefficients of variation of less than 8.3% and 11.4% respectively. The mean concentrations of zonisamide in breast milk and plasma were 9.41 +/- 0.95 and 10.13 +/- 0.45 mu g/mL, respectively. Antibacterial Spiropyrimidinetriones with N-Linked Azole Substituents on a Benzisoxazole Scaffold Targeting DNA Gyrase

Herein, we report spiropyrimidinetriones (SPTs) incorporating N-linked azole substituents on a benzisoxazole scaffold with improved Gram-positive antibacterial activity relative to previously described analogues. SPTs have an unusual spirocyclic architecture and represent a new antibacterial class of bacterial DNA gyrase and topoisomerase IV inhibitors. They are not cross-resistant to fluoroquinolones and other DNA gyrase/topoisomerase IV inhibitors used clinically. The activity of the SPTs was assessed for DNA gyrase inhibition, and the antibacterial activity across Gram-positive and Gram-negative pathogens with N-linked 1,2,4-triazoles substituted on the 5-position provides the most worthwhile profile. Directed nucleophilic and electrophilic chemistry was developed to vary this 5-position with carbon, nitrogen, or oxygen substituents and explore structure-activity relationships including those around a target binding model. Compounds with favorable pharmacokinetic parameters were identified, and two compounds demonstrated cidality in a mouse model of Staphylococcus aureus infection.

UV irradiation of 3-amino-1,2-benzisoxazole isolated in an argon matrix leads to the formation of an amino-spiro-2H-azirine. The amino-spiro-2H-azirine was found to photoisomerize back to 3-amino1,2-benzisoxazole and also to a 1H-diazirine, which isomerizes to a carbodiimide. All the reported species were characterized experimentally by IR spectroscopy and confirmed by comparison with theoretical IR spectra. Synthesis and binding affinities of a series of 1,2-benzisoxazole-3-carboxamides to dopamine and serotonin receptors

A series of 1,2-benzisoxazole-3-carboxamides derived from tertiary cycloalkylamines was synthesized and evaluated for affinity for serotonergic (5-HT3 and 5-HT4) and dopaminergic (D-2) receptors using radioligand binding assays. EFFECT OF NOVEL BENZISOXAZOLE DERIVATIVES AGAINST EHRLICH ASCITES CARCINOMA CELLS IN SWISS ALBINO MICE: CYTOTOXIC AND HAEMATALOGICAL STUDIES

The present study is to investigate the cytotoxicity properties and haematological indices of synthesized molecules (S1-S4) in comparison to the standard drug 5 fluorouracil which reflects the anticancer potentials of the above molecules. Cytotoxicity studies were performed by in vitro MTT (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl teterazolium bromide) assay. Haematological studies were done by drawing blood in the drug and synthesized molecules administered animals by retroorbital method. Haemoglobin, RBC, WBC, lymphocytes, Monocytes and Granulocytes counts were measured. All the above experiments were performed against Ehrlich ascites carcinoma (EAC) in Swiss albino mice. The percentage cytotoxicity and percentage viable cell counts revealed anticancer properties of the synthesized molecules. RBC count indicated protection of cell membrane from destruction and the WBC counts along with lymphocytes, Monocytes and granulocytes also show the immunoprotective characteristics of the novel molecules. Thus it has been concluded that the synthesized molecules (S1-S4) if supported by further molecular studies may have a promising role to play as anticancer agents.

Two aqueous mixtures of cationic and anionic surfactants have been studied by means of conductometry, transmission electron microscopy, and microcalorimetry. Their catalytic effects on the decarboxylation of the kinetic probe 6-nitrobenzisoxazole-3-carboxylate (6-NBIC) were also examined in some detail. The mixtures differ profoundly in the hydrophobic match between both surfactant tails, which is perfect for dodecyltrimethylammonium bromide (DTAB) and sodium dodecyl sulfate (SDS) and poor for hexadecyltrimethylammonium bromide (CTAB) and sodium heptyl sulfate(SHS). This difference is reflected in the more pronounced synergism in critical aggregation concentration and catalytic efficiency of the DTAB/SDS mixture and in the phase behavior of the mixtures. CTAB and SHS can be mixed in a 1:1 ratio without precipitation, forming both small, unilamellar and large, multilamellar vesicles. In DTAB/SDS mixtures, however, precipitation of the catanionic surfactant occurs for a mole fraction of DTAB (x) between 0.3 and 0.7, while both vesicles and large bilayer fragments are formed for x = 0.8. The excess of DTAB in the x = 0.8 mixture results in the solubilization of the Vesicles by DTAB micelles close to the cmc of pure DTAB. A network of interconnected threadlike cylindrical micelles was found as an intermediate stage of aggregation between the vesicles and the mixed micelles. These cylindrical micelles are formed exclusively on further increasing the surfactant concentration.

Depending on the hydrophobic group content of the cross-linked copolymers, a conformational transition was revealed by viscosity measurements which is accounted for by intramolecular micelle formation and intermolecular aggregation. The hydrophobic microdomains of the cross-linked polysoaps were characterized by hypsochromic shifts of the long-wavelength absorption band of methyl orange as a solvatochromic probe, noncovalently bound to the macromolecule. Catalysis of the unimolecular decarboxylation of 6-nitrobenzisoxazole-3-carboxylate by the cross-linked copolymers was investigated in aqueous solution at pH 11.3 and 30 degrees C. The cross-linked polysoaps exhibited higher catalytic activities for decarboxylation than the corresponding non-cross-linked copolymer analogues. A maximum in rate constant was found at about 0.2% (w/w) of cross-linking agent in the cross-linked copolymers.

Multi-pharmacophore Approach to Bio-therapeutics: Piperazine Bridged Pseudo-peptidic Urea/Thiourea Derivatives as Anti-oxidant Agents

In an attempt to develop synergetic, potential and selective anti-oxidant agents, a series of novel piperazine bridged pseudo-peptidic urea/thiourea derivatives (5a-x) are designed by molecular docking study, synthesized and evaluated for their in-vitro anti-oxidant activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and dimethyl-4-phenylenediamine (DMPD) methods. The results revealed that compounds 5q, 5s, 5w and 5x are good anti-oxidant agents. Amongst, compounds with aromatic side chain, thioureal fragment with electron donating group-substitution, particularly, 5s and 5w are the most active members exhibiting remarkable anti-oxidant property. The docking results are complementary to the experimental results. The correlation study revealed that compounds derived from phenylalanine and tryptophan containing electron donating group are excellent antioxidants. [GRAPHICS] .
